No, just create an alias to the folder that the .exs files are in and put it in your new valid Sampler Instruments folder. Then, in Options make sure that EXS will look on the proper volume (Local or External) for the samples. Then, each time you select an instrument in EXS, it may take a second to find the samples, but once it does the instrument will load quicker.
