The installers linked on http://glade.gnome.org/ are not
only a wee bit outdated, they do some funny business with
the PATH envvar IIRC (which is strongly discouraged). That
last bit is most likely the cause of the errors you are
seeing, unless you installed from some other means which
you did not specify ;)

So currently, the easiest way goes like this:
1) download glade3-bundle_3.8.0-1-20110405_win32.zip [1]
    development of that is tracked on [2], btw
2) extract glade3-bundle_3.8.0-1-20110405_win32.zip
    to some_location
3) execute some_location/bin/glade-3.exe

And before somebody asks: I'm waiting on the next GTK+ 2.24
bugfix release which will hopefully have the win32 specific
fixes from the gtk-2-24-win32 branch [3] applied before
building a new bundle containing glade3-3.8.1 [4] :)

     (mostly) complete stack usable with MinGW's gdb, so
     even win32 users can start generating backtraces when
     requested...
