The mouse can behave oddly, 2.6 still doesnt detect my mouse behind an 
input-switch properly, and when dual-booting between 2.4 and 2.6 you'll 
notice that 2.6 does more mouse-acceleration internally.

The responsiveness is significantly improved with running applications. You 
have to realise though, that if you dont use or touch an application, the 
kernel _really_ gives it a low priority and swaps it out. When leaving the 
machine for a few hours and coming back it takes many seconds for the kernel 
to realize you are using the input layer again (it is like typing  through a 
ssh-tunnel with a 1 second ping).

Also I've had a few "hangs" where the computer is unresponsive for a minute a 
so. A similar bug-report was posted against konqueror, so I am not the only 
one seeing it, but it happens very rarely.

All in all mostly good, but different.
