Last week there was a debate about this as well.  Tom Allison is
building something like that, on the moment, using HTML::FromMail
and IMAP.

Keep in mind that MailBox is meant to do e-mail batch-processing,
so your performance may be less that you wished for.  I have a
developed a fast cluster optimized folder type recently based on
MailBox, however that isn't open source/free (yet)

Maildir is the best choice, presumably.  You will need to implement
some "keep folder open" caching, but it is very well doable.
