You can't easily. While you might see some users running long bug
queries right now based on server load or alike, you won't be able to
see e.g. users sitting in front of their screen, writing a bug comment
for the last 3 hours and sending that a few milliseconds in the
future. Right after you checked for long running queries depending on
the load. Bugzilla is stateless web tech, there's not much to check
for.

You need to focus on making restarts somewhat fast, scheudling
maintenance windows and otherwise simply do it. Most users might not
even notice, depending on how fast your hardware is. Some workarounds
are concepts like simply not hosting on bare-metal, but some VM and
only restart the VM, which might take far less time than running
hardware BIOS and stuff. Live patching gets more and more common in
modern OS as well, so that one might not need to restart too often as
well.

Don't overcomplicate things, though.
