Personally, I use cygwin's rsync to mirror profiles and data from my 
workstation to my laptop and vice versa. You just need to make sure to 
sync in the proper direction before doing any work or downloading mail 
to local folders. I'm not sure about fat32 since I use ntfs on both 
machines.
