I asked in the Cygwin mailing list and the answer suggested that Cygwin Python should be able to support WinDLL technically but it has not been implemented yet. I'm helping out with the perl Ctypes equivalent, and cdecl, stdcall and both 64bit fastcalls do work perfectly there.
