Triangular lattice holes in slab

Dear sir,

         For triangular lattice holes in slab structure I have created a
linear waveguide in the middle of the 5*5 structure. Now to use this as a
biosensor to detect cancer cells, should I replace all the holes with RI =
1 (air) by the RI value of the cancer cell?
