Ok, my bad. The standard ssh service is defined with 'TCP & UDP'. Go back to
your custom service and change/edit to set 'TCP & UDP' too.
Or you define another custom service ssh with TCP only to use it as internal
destination service.

You also have to enter the internal destination IP. Here you can define a
custom adress (like your custom service) and use this custom address as
destination.

No, there is no delay. As soon the rule is saved/added it is active (created
as iptables rule in the background).
