Hi all,
same problem with my installation (Win7, 64bit, MikTEX portable 32bit)

@Ulrike:
I tried your steps, they do not help.

I guess that something with the general settings went wrong, because of
several messages regarding blabla.ini file missing.

In my setting:
- pdftex does not work at all
- xelatex does work only with standard settings (koma-script, fancybox and
all the other nice packages does not work - the system seems to does not see
the necessary files

A complete new download + installation does not fix my problem.
Any ideas?

Well I will avoid updates in the next days ;-). 
I suggest 
1. Update FNDB as user an admin
2. Synchronize the repository / switch it
3. Recreate the formats

If the problem persist: make a bug report. I have seen already too
other messages which indicates that something is wrong.
