Nothing will compile

I run MikTeX 2.9 on Win10, everything kept up to date with the Update
(Admin) function. Something has happened recently -- I don't know what
-- and files that used to compile perfectly no longer work. They do
work on another computer I have with the same MikTeX setup.
When I try to typeset this:

% !TEX encoding = UTF-8
\documentclass[12pt]{article}
\usepackage{fontspec}
\usepackage{xunicode}
\usepackage{xltxtra}
\setmainfont{Cardo}
\begin{document}
\noindent A little bit of text here; nothing special.\\
And another line here. \\
\addfontfeature{Numbers=OldStyle}0123456789
\end{document}

I get this error message:

The Cardo font is installed on my system and works in other applications
(and used to work fine in TeX). Any suggestions?
