Access from two computers

I am looking for the most efficient way for accessing an ecco file from two computers on a wifi network. A USB key on my router contains the file. However I cannot access it on a computer if it is open on the other one. I suppose one approach is to never leave a computer while the file is open. Ecco has a complex way of synchronizing files. I had used it many years ago, but if I remember correctly it slowed down the system because it constantly updated files. It is also not simple to set it up. I have also used Dropbox, but then the back-ups would only be functional on one of the computers, or in another Dropbox folder. Can you please indicate if there is a simpler way?
