Once again, you misunderstand how gettext works. *Do* read the 
documentation I pointed you to before asking questions, it explains 
gettext concepts pretty well.

You can't, because it doesn't make any sense. You can create such .po 
file in any text editor, but why would you want to do it when running 
xgettext is trivial (and, I believe, demonstrated in gettext docs) is 
beyond me.
