This should be possible, and some of the LNX-BBC developers have been
interested in doing it.  (There's another approach in which you would
copy the iso9660 image over HTTP or TFTP into a RAM disk, rather than
NFS mounting it -- this would be slower at boot but faster to use
afterward, on client machines with at least 70 MB or 80 MB of RAM.)
Instead of modifying an existing root.bin, you should compile the
whole distribution from source code (and then you can choose exactly
what goes into root.bin and edit the scripts which live inside of it).
