Emojipedia has 512x512 pixel transparent-background PNG renderings of 
many emoji designs, including the open source Google Noto Color Emoji. I 
find it a most useful resource.

All letters are available as regional indicator symbol emoji:

I use and recommend Inkscape for creating icons. I make a 512x512 pixel 
document and export to PNG with an opaque or transparent background. You 
can obtain letters in any font, of any size or weight or colour, using 
the Inkscape text tool. I do not think Inkscape supports emoji fonts but 
that will not be a problem if you only want letters.
