I had some trouble with this too.  I use the tftpd32 program instead of
the Microsoft TFTP, so maybe it's not exactly the same issue you're
having but what I noticed was the path was /pxes/config, with the
leading root slash.  So as counter intuitive as it seems I moved the
folder off the root and it worked.  I'm pretty sure if it's properly
configured you don't need to do that, but it got me to where I could at
least play with things.
 
I believe there's a ftp setting for 'virtual root' to allow it to ignore
the leading /, but since it works I'm content to move to the next
barrier I'm trying to get over.
 
It looks to me like pxelinux.0 loads, then it reads
pxelinux.cfg/01-90-12-02-50-2e-99 (or whatever the mac of the machine
is) for the machine config file.  Note this is reletive to the tftpd
folder (not root).  Then you select what boot option you want.  Then it
loads pxes-0.8-9PB.initrd (or whatever you selected).  Then it loads
/pxes/config/default.conf (note relative to root).
 
That's what was messing me up, my mac config file had to go into
/tftpdroot, but the default.conf for the initrd image goes in
/pxes/config off the root.
 
This may not be your problem, but it's something I stubled over.  Hope
this helps.  I'm pretty sure with a properly configured virtual root
setting this wouldn't happen.

I have given the path to the directory containing the boot files. If I
use MS tftp, do i need to delete this entry? 
I copies the files to the root of tftp, but my Pxes client still says
PXE-T01:File not found and PXE-E3B:TFTP Error- File not found. What
wrong am I doing?
I have followed all the steps outlined in Pxes-MS only environment
except using MS tftp server.
Is pxelinux.0 is the file to be loaded by the client?
