 That's not the way you'd want to do it, although it's possible.

 If you change what /usr/bin/python points to, then yum won't be the
only thing that doesn't work. And any update to "python" will revert
the change.
 What you want to do is change your script that needs python-2.6 to
point to /opt/python or whatever ... or ever better install python26
from EPEL, and point your script to /usr/bin/python2.6
