That will depend on how you are doing the greylisting. I believe it was 
greylite, but i'm not positive, that would implement greylisting with a 
kind of drop in replacement for rblsmtpd. If that's the case, you should 
verify with the greylite author, which apparently has turned greylite 
into a comercial solution.

If you're using a patched version of qmail-smtpd, you just need to use 
plain old rblsmtpd to set it up. Take a look at 
http://www.lifewithqmail.org/lwq.html#rblsmtpd.
