You could add your change to /etc/rc.d/rc.local
so that it changes at the end of boot up,
but there may be something else in the system which
is strange. My FC5 shows:

# ll /dev/ttyS0
crw-rw---- 1 root uucp 4, 64 Jul  3  2006 /dev/ttyS0
