Sorry to hear you are having trouble with the VR-500 failing to work with both tk500 and ADMS software when it had worked OK in the past. Try these fixes: 1) make sure your VR-500 is not in the PRESET mode (see the VR-500 instruction manual to learn about PRESET mode). Some Yaesu (Vertex) receivers (e.g., the VR-120) will not communicate with a computer when the radio is in the PRESET mode. 2) install new alkaline batteries in the VR-500. Do not use an AC power adapter. 3) make sure no other software is using the COM port (e.g., PDA synchronization software). Some PDA software captures the COM port even when the software is not executing. 4) make sure you are not trying to write invalid frequencies into the VR-500.
