 From what I understand, and, I am far from a Mac user, is that the the most 
recent versions of both PHP and GTK have been worked on to compile on Mac, and 
GTK so to build against it natively without Xorg. However. I know crap about it.

Last I actually knew, the user has to run a crap load of Linux utilities, 
including XOrg, to run GTK2. Let's see what Anant has to say, he is the only one 
I know of who would actually know for sure. ;)
