The issue isn't to do with your upgrade to MiKTeX 2.9, it's to do with your upgrade to Windows 7 (presumably you were previously using Windows XP?). In Windows Vista and Windows 7, unlike in Windows XP, the default permissions on the Windows root drive (C:) don't allow creation of files (only folders). If you want that command to work without using -o (as in Ulrike's email) then you'll have to use an elevated command prompt but I wouldn't recommend it.
