The pref is opera:config#UserPrefs|LevelOfUpdateAutomation which would be set to 0 to disable auto update checking. So you could put the following either in /etc/operaprefs_default.ini or (if you wanted to use the custom settings as per my previous email) in /usr/share/opera/custom/defaults/operaprefs.ini.
