Ken,

None of the changes you mention should pose any problem.

I have two Toshibas (but not your model) along with several other, mostly handmedown, laptops and I swap hard drives among them fairly regularly.  I do have some occasional problems swapping, but if you're going to install using the same machine that the drive will live on, these are irrelevant.  

Blasting an updated BIOS is probably a good idea but also probably not germane to your question.  Unlikely that BIOS version will be relevant to the size hard drive the machine can accomodate.
    In my case, I got a handmedown 5101-S101 Toshiba that was getting uncontrollably hot and thereby frying both batteries and hard drives.  (Which was the reason I got the box ... for free).  A bit of google research showed that the BIOS had a mismatch to CPU and the fans weren't turning on.  And a BIOS upgrade fixed the problem.  

I've not run any drives .. quite .. that .. big.
