You'd have to hook in to a lot of functions, like displaying messages to the screen and processing user input, etc. Other scripts that rely on nicks (like friends.pl f.e.) would probably still use the real nick. I don't think it's possible to 'overwrite' the internal list of nicks irssi gathers from the irc server...
