There are three different problem in your post.
A: There is no command line option called "-connect", you need to use 
"-config" to launch the viewer with a .VNC file.
B: You created the VNC file manually, and it the format is not correct.
C: You are not specifying the file extension ".vnc".

Follow these steps to get create a working .VNC file; which will solve 
problem B.
Double-click on UltraVNC Viewer and connect to "mlmk1" manually.
Right-click on the vncviewer icon in the Windows taskbar to bring up the 
menu.
Choose "Save connection info as..." to create a new .VNC file.
The viewer will ask if you want to save the password in the file; choose 
yes. 

Lets assume the filename you picked in step 4 is "mlmk1.vnc".  Your 
command line should be as follows; which takes care of A and C.

     "C:\Program Files\UltraVNC\vncviewer.exe" -config mlmk1.VNC

The .VNC files may already be associate with VNCviewer if you picked that 
option during installation. 
If so, you should be able to double-click on a .VNC file to launch the 
viewer.

Pardon my ignorance but the online documentation states:
Connecting VNCViewers:
-connect host[:display]

My mlmk1.vnc contains:
"c:\Program files\ultravnc\vncviewer.exe" -connect mlmk1

mlmk1 is defined in C:\WINNT\systems32\drivers\etc\hosts

I can still access mlmk1 by double-click on VNCViewer icon.

Any hints ?
