Once upon a time there were HTML image maps.

Programs as KImageMapEditor or gimp could help you to built them.

Since you'll need JS anyway I'm not even sure if they could be the most 
efficient way to achieve your target, but somehow you could start from 
HTML image maps to have at least a fall back alternative to a complete 
JS solution.

A more modern approach could possibly be to use the HTML5 canvas.

Clickable maps are a common problem for web programmers and there should 
probably be some examples and some library.
