I agree, there is no writing rules about how to use SCRIPTDIR but if you look
at
the usage in bsd.ports.mk, it is supposed to host script that overwrite exiting
targets or scripts from Tools/scripts and run automatically by bsd.ports.mk.
If you except a couple of ports such as japanese/zipcodes scripts are usually
run from FILESDIR. And yes, you don't need to copy the scrip before running it

So this is fine to me:
MAKE_CMD=${SH} ${FILESDIR}/xvfb-run.sh ${GMAKE}

You can submit a new patch, I'll try add this port before the end of the week.
