From the sound of it, I'm not going to find this new "empty" option on
my current version. Time to plan the upgrade...
 
I agree with Jorge about regular expressions, and vote for a checkbox
for string and numeric values. To keep the UI simpler, it could only be
displayed on the advanced search page. 
 
Am I correct in guessing that there is no query mechanism for issues
with a blocked by/related to/duplicate of relationship?

If you mean "empty" as an option, yes we have it.
I think I did it myself in SCB368... I guess that was the development I
mentioned before :-(

I don't think regular expressions are the easier approach to a regular
user (though it's definitively the most powerful). 
And we still have another issue related (unassigned 'user attributes')
